Weinstein, the onetime Tinseltown powerbroker whose films earned dozens of Oscars, was charged with rape and another sex crime in NY late last month, almost eight months after the first public allegations against him surfaced.
This comes seven months after several media outlets published reports of alleged abuse by the movie mogul.
"Mr. Weinstein has denied these crimes".
"As awful a crime as rape is, it is equally reprehensible to be falsely accused", he said.
Weinstein was not expected to say much but this was still a momentous day in downfall of a man who dozens of women have accused of sexual misconduct in what is likely to become the signature trial of the #MeToo movement.
Vance Jr. announced last week the grand jury handed up charges of rape in the first and third degrees, as well as committing a criminal sexual act in the first degree.
Other actresses who have publicly accused Weinstein of sexual misconduct include Uma Thurman, Salma Hayek and Cate Blanchett.
Wearing a black suit and tie, Mr Weinstein and his lawyer Benjamin Brafman walked past a crowd of reporters and photographers as they arrived at the courthouse in Manhattan. Prosecutors say Weinstein held an unnamed woman in a room against her will and raped her.
The film will be called Predator, and De Palma, who is best known for iconic films such as Carrie, The Untouchables and Scarface, has said the character will be inspired by Weinstein, but will have a different name. He was also ordered to wear a Global Positioning System monitoring bracelet and surrender his passport and is only allowed to travel within NY and CT.
Brafman also said he thought the unnamed victim could come out after he filed subpoenas.
"Melissa did not learn that Brafman was actually then or would later be Weinstein's lead criminal defense attorney until after turning over the video as evidence".
Weinstein is due back in court in September.
